The proposed research aims to tackle significant challenges in the field of ophthalmology, particularly concerning corneal endothelial dysfunction and the global shortage of human donor corneas. The innovative and tailor-made therapeutic strategy intends to fabricate an artificial Descemet membrane composed of the constituents of the natural Descemet membrane using 3-D printing technology, and to grow a monolayer of corneal endothelial-like cells onto this substrate obtained by differentiating human adipose-derived cells. Thus, the project seeks to provide a sustainable and accessible alternative to conventional corneal transplantation methods reliant on corneal endothelial tissue donors. The objectives of the TECE project are ambitious and go beyond the current state of the art in several ways: - Novel concepts and approaches: The project proposes the fabrication of a monolayer of corneal endothelium cells and an artificial Descemet membrane using innovative techniques such as 3D bioprinting, stem cell differentiation and tissue engineering. These approaches represent novel concepts in the field of corneal tissue engineering and offer potential breakthroughs in the treatment of corneal endothelial dysfunctions; - Interdisciplinary development: TECE integrates knowledge and methodologies from various disciplines, including (bio)chemistry, physics, materials science, molecular biology, and medicine. By combining expertise from these diverse fields, the project aims to develop comprehensive and multidisciplinary solutions to address the challenges posed by corneal endothelial dysfunction, particularly in maintaining the functionality of corneal endothelial-like cells and the transparency of the artificial Descemet membrane; - Translation of research into clinical practice: The ultimate goal of the TECE project is to translate research findings into clinical applications, providing patients with access to innovative and personalized treatments for corneal disorders. To achieve this, the project is structured to align with existing surgical techniques, ensuring wider acceptance by the medical community. This ambitious objective requires bridging the gap between laboratory research and clinical implementation, thereby advancing the field of regenerative medicine and biotechnology in ophthalmology. In summary, the objectives of the TECE project demonstrate a commitment to addressing important challenges in the field of ophthalmology while also pushing the boundaries of current knowledge and technology. By developing novel concepts and approaches and integrating interdisciplinary development, the project aims to provide innovative solutions to improve patient outcomes and address the global shortage of human donor corneas.
