Summary

In addressing this complex challenge of BC bone metastasis, the METABONE project identifies two primary obstacles: i) the lack of a physiologically relevant model that accurately simulates the bone metastatic environment (e.g., the bone tissue and interaction with BC cells), and ii) the ethical concerns associated with current research methodologies. METABONE innovative solution proposes the development of a cutting-edge 3D model using off-the-shelf liquid capsule than can be used on-demand as bioreactors. This model aims to closely mimic the bone niche microenvironment and the colonization of BC cells into the niche, allowing for more accurate studies and potential drug testing. Additionally, we introduce robotic micro-automation to create the 3D model, enhancing the precision and reproducibility of the experiments, significantly reducing human error and the need for animal models. METABONE objectives are set to push the boundaries beyond the current state of the art, incorporating novel concepts, and interdisciplinary approaches. METABONE intends to: - Model the complexity of bone metastasis by creating a more physiologically relevant model of bone tissue, providing an environment that closely replicates human bone for the study of cancer cell interactions, tumor growth, and metastasis. - Create a scalability and high-throughput screening approach to significantly accelerate the pace of drug discovery and development. - Create an ethical and efficient model to reduce the reliance to animal testing models, by providing a more accurate alternative to streamline the drug discovery process, making it more efficient and less costly. - Invest in novel concepts by integrating robotic micro-automation to enhance accuracy, efficiency, and reproducibility on the creation of the 3D BC metastasis bone model; and off-the-shelf liquid capsules as versatile on-demand bioreactors for modeling bone metastasis, a significant departure from traditional 2D and animal models. - Invest in interdisciplinarity with the fusion of disciplines, integrating biotechnology, diseases modelling, and robotics. This interdisciplinary approach facilitates the development of novel methodologies for studying BC cancer metastasis and screening drugs, contributing to advancements across these fields. - Create a comprehensive database of research findings to unlock new insights into metastasis mechanisms and treatment, with the use of machine learning algorithms. This data-driven approach exemplifies the project's potential to advance scientific discoveries and societal well-being. The success of this project could set a new standard for modeling various types of cancer bone metastases (e.g., pancreas, liver, melanoma, among others) potentially leading to groundbreaking discoveries in treatment and prevention strategies. The adaptability of this 3D model and robotic technology to other cancer types illustrates the project's potential for widespread impact, offering a versatile tool in the fight against bone metastasis events.
